Introduction of Research Papers using PrimeSurface™
The tumor suppressor gene FBXW7 inhibits cancer stem cell properties, and inhibition of KCa1.1 (calcium-activated potassium channel) is involved in its function; however, the detailed transcriptional regulation of FBXW7 and intracellular signaling induced by KCa1.1 inhibition remain unclear.
Here, we introduce a paper published in the International Journal of Molecular Sciences titled "Transcriptional Up-Regulation of FBXW7 by KCa1.1 K+ Channel Inhibition through the Nrf2 Signaling Pathway in Human Prostate Cancer LNCaP Cell Spheroid Model."
This study used a 3D spheroid model of human prostate cancer LNCaP cells and revealed that KCa1.1 inhibition promotes the transcriptional activation of the tumor suppressor gene FBXW7 via the Akt-Nrf2 signaling pathway.
PrimeSurface® 96 U plate was used to form 3D spheroids for gene and protein expression analyses and other assays showing significantly increased FBXW7 expression and reduced Nrf2 nuclear translocation after KCa1.1 inhibitor PAX treatment. Furthermore, siRNA-mediated suppression of FBXW7 expression was observed.
These results suggested that FBXW7 plays a crucial role in transcriptional regulation. These findings suggest the potential of FBXW7 as a novel therapeutic target.
